I don't think it matters whether you are left or right handed - 
 
I began holding the harmonica in my right hand - it felt more natural.  
Played it this way for 20 or more years. Then I heard Joe Filisko talk on  
amplified sound about 5 years ago. What he said made sense in sculpting the  sound. 
 
I wanted add the huge amplified sound to my arsenal, so spent the time  
needed on learning this most efficient technique, which called for holding the  
harmonica in my left hand. 
 
It took about 9 months before it felt comfortable. Learning how to relax  the 
grip while still keeping an airtight seal was a turning point.
 
I still play about 75% right handed - more of a clean and controlled sound  - 
even "pretty". This is the inner musical voice in me trying to get out.  
However, when I want to get down and dirty, the harmonica switches hands and my  
grip changes. It's like having two options instead of one.
 
Like TB and single note playing, how you hold the harmonica can be more  than 
one way.
