Hi,
If you're playing a c standard-tuned diatonic, and you are playing in
twelfth position, you'd be playing in some permutation of f. 
It's instructive because the twelfth position insists we learn the bends on
the two and three draw holes.  And..., there's the six over-blow, required
if you want to do an f-major scale all the way up the harmonica. 
If you go to youtube and search for twelfth position/Happy Birthday, you'll
probably find a video that Buddha posted a few months ago which is pretty
convincing evidence as to why you'd want to learn twelfth position. 
 Never say never!!
