Well,
I much prefer playing (or singing) standing up. It is easier to get that  
"column of air" vibrating.
For the maximum tone, either playing harmonica or as a vocalist!
Over the past few years, I've been playing with a 25 piece orchestra. The  MD 
requires that I play sitting down, except when soloing.
So I do what I'm told! But for optimum tone, standing up is better.
