If you were to take a listen to some Madcat(especially with Sky King),
Norton Buffalo(especially his 2 early solo albums), "Yee Haw Factor" from
the Flecktones, "Crew Slut" from Frank Zappa and others, you would find
some very creative high-end 2nd and 3rd position diatonic harmonica work.
The truth is that the possibilities are endless.

Because of the physical layout of the draw notes, a standard diatonic can
be thought of as two distinct instruments: holes 1-6 and holes 7-10. Each
has it's strengths and weaknesses and the interface between them can be
tricky. But like everything else, with practice, the two parts can become
one.

The real limitations are nearly always in our imaginations.
